Probiotic Characteristics of Bacillus tequilensis JBC17126 Isolated from Korean Traditional Soybean Paste

Abstract
This study examined the probiotic characteristics (acid tolerance, bile tolerance, antibacterial activity, and adhesive property) of Bacillus strains isolated from Korean traditional soybean pastes. The results of the acid and bile tolerance tests showed that the JBC17126 strain had high viability with 4.32 log CFU/mL of 7.58 log CFU/mL (57.04%) under acid conditions (pH 2.5) and 4.41 log CFU/mL of 7.43 log CFU/mL (59.34%) under bile condition (bile 0.3%). The adhesion assessment for mucin (intestinal mucus) showed that the JBC17126 strain was adhesive to mucin with 5.87 log CFU/mL of 7.49 log CFU/mL (78.37%). The JBC17126 strain, which showed acid tolerance, bile tolerance, and adhesive property, also inhibited Bacillus cereus and Staphylococcus aureus. This strain was confirmed to be Bacillus tequilensis by 16S rRNA gene sequencing, and was named B. tequilensis JBC17126. Based on these results, B. tequilensis JBC17126 is expected to be a useful probiotic with excellent survival and adherence in the intestinal environment.
